Abstract: | In the present study, goat-lung was decellularized and used as a scaffold to support cellular attachment and proliferation. The biological properties of decellularized goat-lung scaffold were enhanced by modifying it with collagen and gelatin, and the effect of modification was evaluated on the growth of human skin-derived mesenchymal stem cells (S-MSCs). MTT assay demonstrated good biocompatibility, cell adhesion and proliferation of S-MSCs over collagen and gelatin modified scaffolds as compared to unmodified decellularized scaffold. Scanning electron microscopy observation confirmed the distribution of S-MSCs in the gelatin and collagen modified decellularized scaffold. Hematoxylin and Eosin staining further revealed that collagen and gelatin modified decellularized scaffold maintained good biocompatibility with S-MSCs. Expression of Keratin18 gene, along with CD105, CD73 and CD44, by S-MSCs over collagen and gelatin modified decellularized scaffold signifies that the cells are viable, proliferating and migrating, and have maintained their critical cellular functions within the scaffold. All the above results suggests that modification of goat-lung decellularized scaffold by collagen and gelatin enhances the growth and proliferation of S-MSCs, and therefore, collagen and gelatin modified decellularized scaffold could be a potential candidate for bio-artificial skin tissue engineering applications. © 2013 American Scientific Publishers, All rights reserved. |
